Although there are plenty of ways and hero combinations to be used for this challenge, there will always be a certain set of heroes more suited for completion than others. The Close Quarters challenge is one example of this as, in this mode, enemies can only be killed when others are nearby. You'll be relying on your team more than ever.

You're definitely going to want Roadhog — there's no question on that one. The entire challenge is centered around getting your targets close enough to your team for your shots to register. Hog's hook is the epitome of useful here.

Next, you'll need heroes with a high damage output so you can bury your enemies six feet under in as short a span as possible. Reaper would do well here, as would Genji with his already close-combat melee swordsmanship. You might also get some use out of Mei with her crowd control capabilities if you're looking to isolate targets one by one.

Winston is a decent choice here, too. His ultimate is entirely close-combat thrashing, after all.

This is also the mission where Reinhardt is most viable. His charge can easily bring you in close for some massive advantage and leave room in your wake for your team to fill in behind.
